RESUMEN

Substantial investments into laboratories, notably sophisticated equipment, have been made over time to detect emerging diseases close to their source. Diagnostic capacity has expanded as a result, but challenges have emerged. The Equipment Management and Sustainability Survey was sent to the Veterinary Services of 182 countries in mid-2019. We measured the status of forty types of laboratory equipment used in veterinary diagnostic laboratories. Of the 68,455 items reported from 227 laboratories in 136 countries, 22% (14,894/68,455) were improperly maintained, and 46% (29,957/65,490) were improperly calibrated. Notable differences were observed across World Bank income levels and regions, raising concerns about equipment reliability and the results they produce. Our results will advise partners and donors on how best to support low-resource veterinary laboratories to improve sustainability and fulfill their mandate toward pandemic prevention and preparedness, as well as encourage equipment manufacturers to spur innovation and develop more sustainable products that meet end-users' needs.

Making user interaction with laboratory equipment more convenient and intuitive should promote experimental work and help researchers to complete their tasks efficiently. The most common form of interaction in current instrumentation is either direct tactile, with buttons and knobs, or interfaced through a computer, using a mouse and keyboard. Scripting is another function typical of smart and automated laboratory equipment, yet users are currently required to learn bespoke programming languages and libraries for individual pieces of equipment. In this paper, we present two open-source, novel and intuitive ways of interacting with and scripting laboratory equipment. We choose the OpenFlexure family of microscopes as our exemplar, due to their open-source nature and smart control system. Firstly, we demonstrate 'OpenFlexure Voice Control' to enable users to control the microscope hands-free. Secondly, we present 'OpenFlexure Blockly' which uses the Blockly Visual Programming Language to enable users to easily create scripts for the microscope, using a drag and drop Web interface. We explain the design choices when developing these tools, and discuss more typical use cases and more general applications.

Specialised mass spectrometers such as those used in noble gas geochemistry are often controlled by bespoke computer systems that get harder to maintain as, along with their expensive interface cards, they become obsolete. This work explains how bespoke interface cards and obsolete versions of software can be replaced with low-cost single board computers and open source software. Although tailored to an ultra-high vacuum helium extraction line the basic elements and microservices detailed in this paper provide a template for controlling a wider range of instruments. •Moving from obsolete computer hardware to current hardware.•Moving from licenced proprietary software to opensource software.•Automating processes to reduce process time and manual steps.

ABSTRACT Objective This study aimed to compare the hematological parameters released by hematological analyzers with those released in customer reports. Methods We conducted a descriptive study in the laboratories of a medium-sized municipality in the state of Minas Gerais registered in the National Register of Health Establishments. Interviews were conducted using a questionnaire to obtain information regarding the parameters released by the analyzers and those available in the customer's report. Results Sixteen laboratories were evaluated, and none of them released all the parameters obtained from the hematological analyzers to customers. The red blood cell distribution width was released in 88% of the laboratories, atypical lymphocytes in 70%, mean platelet volume in 50%, platelet distribution width and platelet count in 20%. No laboratory released information on reticulocytes, fraction of immature reticulocytes and immature granulocytes, nucleated erythrocyte count, immature platelet fraction and reticulocyte hemoglobin, and large platelet rate. Conclusion All evaluated clinical analysis laboratories had at least one parameter that was not released in the customer's report despite being released by the hematological analyzers. The lack of knowledge on the part of professionals about the clinical importance of each parameter of the complete blood count results in a loss in patient assessment, and it is important to include these parameters in the complete blood count report.

Organic electrosynthesis is an increasingly popular tool for driving and probing redox reactions. Recent advances in this field often employ an electrocatalyst to enhance the selectivity and efficiency of electrochemical reactions. A laboratory experiment was developed to introduce students to relevant mechanistic techniques in electrochemistry for analysis of electrocatalytic reactions using aminoxyl-catalyzed alcohol oxidation as a case study. This lab activity employs cyclic voltammetry for qualitative assessment of catalytic turnover prior to introducing students to chronoamperometry, an underutilized technique that facilitates quantitative determination of the rate of catalysis. Students identify and rationalize the important features of reversible electron transfer and a catalytic reaction in a cyclic voltammogram, probe the origin of scan rate effects on these traces, and calculate turnover frequency using a series of chronoamperograms. The method employs safe and readily available reagents: basic aqueous buffer solution, alcohol substrate, and an inexpensive organic aminoxyl catalyst. Student data presented herein were obtained from a course attended by undergraduate students, graduate students, and pharmaceutical chemists.

Patient care and public health require timely, reliable laboratory testing. However, clinical laboratory professionals rarely know whether patient specimens contain infectious agents, making ensuring biosafety while performing testing procedures challenging. The importance of biosafety in clinical laboratories was highlighted during the 2014 Ebola outbreak, where concerns about biosafety resulted in delayed diagnoses and contributed to patient deaths. This review is a collaboration between subject matter experts from large and small laboratories and the federal government to evaluate the capability of clinical laboratories to manage biosafety risks and safely test patient specimens. We discuss the complexity of clinical laboratories, including anatomic pathology, and describe how applying current biosafety guidance may be difficult as these guidelines, largely based on practices in research laboratories, do not always correspond to the unique clinical laboratory environments and their specialized equipment and processes. We retrospectively describe the biosafety gaps and opportunities for improvement in the areas of risk assessment and management; automated and manual laboratory disciplines; specimen collection, processing, and storage; test utilization; equipment and instrumentation safety; disinfection practices; personal protective equipment; waste management; laboratory personnel training and competency assessment; accreditation processes; and ethical guidance. Also addressed are the unique biosafety challenges successfully handled by a Texas community hospital clinical laboratory that performed testing for patients with Ebola without a formal biocontainment unit. The gaps in knowledge and practices identified in previous and ongoing outbreaks demonstrate the need for collaborative, comprehensive solutions to improve clinical laboratory biosafety and to better combat future emerging infectious disease outbreaks.

Accessibility to potentiostats is crucial for research development in electrochemistry, but their cost is the principal drawback for their massive use. With the aim to provide an affordable alternative for resource-constrained communities, we present a low-cost, portable electrochemical workstation that integrates an open-source potentiostat based on Arduino and a smartphone application. This graphical user interface allows easy control of electrochemical parameters and real-time visualization of the results. This potentiostat can perform the most used electrochemical techniques of cyclic and linear voltammetry and chronoamperometry, with an operating range of ±225 µA and ±1.50 V, and results that are comparable with those obtained with commercial potentiostats. Three applications reported here demonstrate the capacity and the good performance of this low-cost potentiostat as a teaching tool: identification of redox pairs, electrochemical characterization of pencil graphite electrodes, and detection of heavy metals using an electrodeposited film of bismuth on the pencil graphite electrode. Furthermore, detailed schemes of the device and its software are entirely available, expecting to provide an open-source potentiostat easy to replicate to further support education in electrochemical fundamentals and instrumentation.

Electrophysiological recordings lead amongst the techniques that aim to investigate the dynamics of neural activity sampled from large neural ensembles. However, the financial costs associated with the state-of-the-art technology used to manufacture probes and multi-channel recording systems make these experiments virtually inaccessible to small laboratories, especially if located in developing countries. Here, we describe a new method for implanting several tungsten electrode arrays, widely distributed over the brain. Moreover, we designed a headstage system, using the Intan® RHD2000 chipset, associated with a connector (replacing the expensive commercial Omnetics connector), that allows the usage of disposable and inexpensive cranial implants. Our results showed high-quality multichannel recording in freely moving animals (detecting local field, evoked responses and unit activities) and robust mechanical connections ensuring long-term continuous recordings. Our project represents an open source and inexpensive alternative to develop customized extracellular records from multiple brain regions.

RESUMEN El Laboratorio de Cirugía Experimental de la Universidad de Ciencia Médicas de Villa Clara cuenta con la tecnología necesaria para el montaje y desarrollo de técnicas y habilidades quirúrgicas y microquirúrgicas, las cuales son utilizadas por los profesionales y estudiantes en la docencia y la investigación biomédica. Se realizaron varias innovaciones con la recuperación de equipos, instrumentos y materiales en desuso; se creó el gimnasio quirúrgico conformado por modelos inanimados sintéticos, y se implementó el modelo «pollo¼, como técnica alternativa al uso de animales vivos en la experimentación animal. Se han desarrollado: importantes proyectos de investigación, entrenamientos de cirugía y microcirugía experimental, diversos programas docentes, y exitosos cursos internacionales con estudiantes y residentes. El laboratorio constituye un valioso aporte de alternativas éticas, económicas y sanitarias, que han favorecido la formación de recursos humanos y un servicio científico-técnico de calidad para la docencia y la investigación.


ABSTRACT The Laboratory of Experimental Surgery at the University of Medical Sciences of Villa Clara has the necessary technology for the assembly and development of skills, as well as, surgical and microsurgical techniques, which are used by professionals and students in teaching and biomedical research. Several innovations were made with the recovery of equipments, instruments and disused materials; the surgical gymnasium was made up of synthetic inanimate models, and the "chicken" model was implemented as an alternative technique to the use of live animals in animal experimentation. Important research projects, training in surgery and experimental microsurgery, diverse teaching programs, and successful international courses with students and residents have been developed. The laboratory constitutes a valuable contribution of ethical, economic and sanitary alternatives, which have favored the formation of human resources and a qualified scientific-technical service for teaching and research.

The article presents information to assess possibility of using on-line questionnaires to obtain statistic data concerning activities of laboratory services in the region. The sets of standardized statistical indicators were obtained for assessing performance of such regional services as the structure of laboratory research; comparative standardized values of availability of examinations per 1000 of population; comparative standardized values of equipment support in the regions of the Russian Federation and employment rates and staffing with medical personnel of laboratories.

O processamento tecidual em cirurgia de Mohs visa à confecção de lâminas histológicas que permitam a análise de 100% das margens cirúrgicas. É uma etapa crítica e passível de erros. Como não há padronização na montagem dos blocos, há desnivelamento das superfícies de corte das diferentes amostras, levando à necessidade de contínuos ajustes no eixo X-Y no interior do criostato, lentificando o processo. Visando à resolução desse problema, desenvolveu-se um dispositivo que minimiza quaisquer inclinações dos blocos, mantendo-se as margens cirúrgicas paralelas em todas as amostras, acelerando-se o processo e mantendo-se a alta qualidade das lâminas histológicas.


The tissue processing in Mohs surgery aims at histological slides that allow the analysis of 100% of the surgical margins. The embedding tissue is a critical step and prone to errors. As there is no standardization when mounting the blocks, there may be unevenness in the different samples cutting surfaces, leading to the need for continuous adjustments on the X-Y axis inside the cryostat, slowing down the process. A device was developed to solve this problem, minimizing any blocks inclination, keeping the surgical margins parallel in all samples, accelerating the process, and maintaining the histological slides high quality

Described is the creation, deployment, and evaluation of a video produced about the synthesis and applications of metal-organic frameworks (MOFs). The goal of this project was to gauge the impact of viewing the video on high school students' conceptions of authentic chemistry practices and applications. Additionally, comparisons were made between the use of the video and more traditional face-to-face presentations given by professional scientists. Observations, student surveys, and an interview with the high school chemistry teacher demonstrated the utility of such a video. Specifically, the students who viewed the video reported learning more about the nature of laboratory work in chemistry than other students who did not view the video. Students, regardless of whether they viewed the video or just received a presentation, reported growth in understandings of the applications of chemistry research and porous nanomaterial. Other research chemists are encouraged to consider ways that they could document on video the research that they are performing in order to introduce an untapped audience (high school students) to authentic chemistry research in a practically simple manner. During times of crisis, such as a pandemic, online videos could be a useful tool for high school chemistry teachers to use in collaboration with research faculty, particularly when schools are closed.

Animal behavioral paradigms, such as classical conditioning and operant conditioning, are an important tool to study the neural basis of cognition and behavior. These paradigms involve manipulating sensory stimuli in a way that learning processes are induced under controlled experimental conditions. However, the majority of the commercially available equipment did not offer flexibility to manipulate stimuli. Therefore, the development of most versatile devices would allow the study of more complex cognitive functions. The purpose of this work is to present a low-cost, customized and wireless-operated chamber for animal behavior conditioning, based on the joint operation of two microcontroller modules: Arduino Due and ESP8266-12E. Our results showed that the auditory stimulation system allows setting the carrier frequency in the range of 1 Hz up to more than 100 kHz and the sound stimulus can be modulated in amplitude, also over a wide range of frequencies. Likewise, foot-shock could be precisely manipulated regarding its amplitude (from ∼200 µA to ∼1500 µA) and frequency (up to 20 pulses per second). Finally, adult rats exposed to a protocol of cued fear conditioning in our device showed consistent behavioral response and electrophysiological evoked responses in the midbrain auditory pathway. Furthermore, the device developed in the current study represents an open source alternative to develop customized protocols to study fear memory under conditions of varied sensory stimuli.

We built a low-cost and hand-held device to image and analyze microfluidic droplets mainly for educational/teaching purposes in laboratory settings of universities. The device was assembled based on a Raspberry Pi with a camera attached on the back and an LCD screen on the top. We evaluated the performance of this device to capture images and videos to visualize high-throughput droplet generation in a microfluidic device. The qualities of imaging resolution and speed were sufficient for us to perform subsequent droplet analysis quantitatively through automatic image possessing. Droplet characteristics including droplet size, volume, and dispersity, as well as droplet intensity, have been measured, showing the potential of this device to analyze droplet-based assays. Most importantly, in addition to learning the knowledge and principles from classroom lectures, students can thus gain practice of using an advanced, state-of-the-art technology in a laboratory course. It will also open up opportunities to train students with skills of interdisciplinary thinking and learning.

ABSTRACT Introduction: The harmonization of equipment is recommended in clinical laboratory practice aiming for the homogeneity of results when similar or equivalent analyzers are used to perform routine testing. Objectives: To conduct a study of equivalence between the biochemical analyzers Labmax 240® (E1) and Labmax 240 Premium® (E2) through the matching results and the statistical value analysis of dosages. Materials and methods: We evaluated tests with glucose, total cholesterol, triglycerides, uric acid, aspartate transaminase (AST), alanine transaminase (ALT) and lactate dehydrogenase (LDH), all with 40 repeated measurements, performed in both equipments. The Clinical and Laboratory Standards Institute (CLSI) EP09-3A protocol was used to conduct the comparison test between E1 and E2 equipment, with subsequent evaluation of the results for statistical analysis determining the Pearson correlation coefficient (r) and indexes comparison error with EP Evaluator® software. Results: Regarding the values of the Pearson correlation coefficient, all tests showed a strong correlation between equipment with r > 0.989, except for the dosage of LDH (r = 0.982). This dosage failed not because the value of r, but due to the values obtained in the error index being larger than the total errors index allowed. Discussion: Compared to clinical criteria, the results of the analyzers are approximately equal, but this control process must be done continuously in order to prevent and track random errors within the laboratory routine. Conclusion: The process of harmonization of multiple devices that perform the same laboratory parameters is essential for ensuring quality and reliability of laboratory results and should be standardized and included in routine clinical analysis laboratories.


RESUMO Introdução: No laboratório clínico, é recomendável a harmonização de equipamentos que visem à homogeneidade dos resultados, quando analisadores similares ou equivalentes são utilizados para desempenho da rotina de realização dos testes. Objetivos: Realizar um estudo de equivalência entre os analisadores bioquímicos Labmax 240® (E1) e Labmax 240 Premium® (E2). Materiais e métodos: Foram avaliados os testes glicose (GLI), colesterol total (COL), triglicerídeos (TRI), ácido úrico , aspartato aminotransferase (AST), alanina aminotransferase (ALT) e lactato desidrogenase (LDH), todos com 40 dosagens repetidas, realizadas em ambos os equipamentos. O protocolo do Clinical and Laboratory Standards Institute (CLSI) EP09-3A foi utilizado para conduzir o teste de comparação, com posterior avaliação dos resultados pela análise estatística no software EP Evaluator®, com determinação do coeficiente de correlação de Pearson (r) e comparação de índices de erro. Resultados: Em relação aos valores do coeficiente de correlação de Pearson, todos os testes apresentaram forte correlação entre os equipamentos, com r > 0,989, exceto para a dosagem de LDH (r = 0,982), que foi reprovada, não em função do valor de r, mas devido aos valores obtidos em relação ao índice de erro, o qual é maior do que os índices de erro total permitido. Discussão: Diante dos critérios clínicos, os resultados dos analisadores são aproximadamente iguais, porém esse controle do processo deve ser feito continuamente a fim de impedir e rastrear erros aleatórios dentro da rotina laboratorial. Conclusão: O processo de harmonização de múltiplos equipamentos que realizam os mesmos parâmetros laboratoriais é fundamental para a garantia da qualidade e da confiabilidade dos resultados laboratoriais, devendo ser padronizado e incluído na rotina dos laboratórios de análises clínicas.

El avance de los estudios moleculares en el cáncer de mama ha proporcionado argumentos para establecer subclasificaciones de importancia diagnóstica y terapéutica, las cuales están acercando a la práctica diaria la deseada posibilidad de una terapia personalizada para las pacientes, al catalogar mejor el tipo de neoplasia. La mayoría de los estudios actuales buscan relacionar fenotípicamente estos subtipos con los factores clásicos reportados en el informe anatomopatológico del cáncer de mama, a través de técnicas convencionales con hematoxilina y eosina. La inmunohistoquímica es una herramienta confiable que permite subclasificar estas neoplasias en diversas categorías usando marcadores, con lo cual se logra establecer parámetros necesarios en el diagnóstico y pronóstico. En el presente trabajo se exponen los principales marcadores inmunohistoquímicos usados en Cienfuegos así como su utilidad.


Molecular studies development in breast cancer has provided arguments to establish subclassifications of diagnostic and therapeutic importance, which are bringing to the daily practice the desirability of a patients personalized therapy, to better identify the type of neoplasia. Most current studies seek to phenotypically relate these subtypes to the classic factors reported in the breast cancer anatomopathological report, using conventional hematoxylin and eosin techniques. Immunohistochemistry is a reliable tool that allows subclassification of these neoplasms in different categories using markers, which can establish necessary parameters in diagnosis and prognosis. In this paper the main immunohistochemical markers used in Cienfuegos are presented as well as their usefulness.

Off-the-shelf computer cooling hardware was used to construct a closed-loop air cooling system (CLACS) that is distinguished by scalability, low energy, and no tap water consumption. Constructed to be generally used with laboratory condensers, the system was tested with several common low and high boiling solvents and showed a condensation performance equivalent to conventional tap water cooling. Reaction yields were therefore unaffected. Also, long-lasting Soxhlet extractions showed no performance loss relative to conventional cooling. Optionally, the device can be assembled from low-voltage components and be powered from a battery, rendering it independent of the main power. Both investment and running costs are minimal, allowing a lab-wide adoption and elimination of the two major drawbacks of commonly employed tap water cooling: waste of drinking water and the risk of flooding.

Objective To solve the problems of clinical laboratory module in mobile medical unit such as miscellaneous and mixed equipment,disordered packages,and weak mobility and responsiveness.Methods The concept of modularization was used for the secondary optimization of clinical laboratory equipment and consumables.The specialized vehicle for clinical laboratory was designed with considerations on the advantages of clinical laboratory shelter in equipment and medical vehicle in mobility,so as the values of clinical laboratory module were enhanced in mobile medical unit.Results Modular and standardized management of clinical laboratory equipment was realized to solve the existing problems of equipment.Conclusion The equipment supportability and mobility are enhanced in mobile medical unit,so that the requirements can be fulfilled for medical support missions at peace time and wartime.

Many research and teaching labs rely on USB data acquisition devices to collect voltage signals from instrumentation. However, these devices can be cost-prohibitive (especially when large numbers are needed for teaching labs) and require software to be developed for operation. In this article, we describe the development and use of an open-source USB data acquisition device (with 16-bit acquisition resolution) built using simple electronic components and an Arduino Uno that costs under $50. Additionally, open-source software written in Python is included so that data can be acquired using nearly any PC or Mac computer with a simple USB connection. Use of the device was demonstrated for a sophomore-level analytical experiment using GC and a CE-UV separation on an instrument used for research purposes.
